چکیده

Pharmacokinetic study of paracetamol was conducted in five female buffalo calves following single i.v. injection of paracetamol @ 40 mg/kg. The drug was detected up to 2 h only in plasma, while it was detected for a longer period of 48 h in urine with a mean concentration of 1.87 ± 0.48 and 10.90 ± 1.14 mg/ml, respectively. The mean distribution half life (t 1/2 α) of 0.11 ± 0.01 h and elimination half life (t 1/2 β) of 0.77 ± 0.09 h indicated that the drug is rapidly distributed as well as eliminated quickly in buffalo calves. The rapid elimination of the drug is due to the higher values of rate constant of drug elimination from central compartment (Kel) of 1.471 ± 0.089 h and total body clearance (Cl B ) of 43.67 3.39 ml/kg/min. This led to the lower value of mean residential time (MRT) of 1.00 ± 0.01 h, indicating that paracetamol remained for a shorter period only in the body of buffalo calves. A higher Vd area of 2.34 ± 0.19 L/kg obtained in the present study suggested good distribution of paracetamol in body fluids and tissues which is further supported by the approximate tissue to plasma concentration ratio (T/P) of 0.59 ± 0.13.

Pharmacokinetics of ceftazidime in buffalo calves following intravenous and intramuscular administration

The pharmacokinetic parameters of ceftazidime, a third generation cephalosporin, were investigated in six buffalo calves after single intravenous (IV) and intramuscular (IM) administration at a dose rate of 10 mg/kg body weight. Ceftazidime concentrations in plasma and urine were determined by microbiological assay. Ceftazidime disposition was best fitted by a two-compartmental and a one-compar...

Disposition Kinetics and Urinary Excretion of Paracetamol Following Intravenous Administration in Buffalo Calves (Bubalus bubalis)

Paracetamol (Acetaminophen), a NSAID is well tolerated and lacks many side effects of aspirin. It is a widely used over-the-counter analgesic, antipyretic and weak anti-in ammatory property. The main mechanism is the inhibition of cyclooxygenase (COX) and recent  ndings suggest that it is highly selective for COX-2 (Henz et al., 2008). The present study was aimed to study the disposition kine...

Pharmacokinetics of ceftriaxone in buffalo calves (Bubalus bubalis) following intravenous and intramuscular administration

Pharmacokinetics of ceftriaxone was studied in buffalo calves (Bubalus bubalis) after single intravenousand intramuscular administration of 10 mg/kg body weight. The drug concentrations in plasma samples weremeasured by high performance liquid chromatography with UV detection. Following intravenousadministration, the drug was rapidly distributed (Cpo: 106.5 ± 9.64 μg/ml; t1/2α: 0.09 ± 0.01 h; V...

Plasma Disposition and Urinary Excretion of Levofloxacin following a Single Intravenous Administration in Buffalo Calves

Disposition kinetics and urinary excretion of levofl oxacin was investigated after a single intravenous dose of 3 mg.kg-1 in male buffalo calves. The drug concentration was estimated in plasma and urine by microbiological assay using E. coli as the test organism. Disposition kinetic parameters were determined using a two-compartment open model and an appropriate dosage schedule was computed. Dr...

Kinetic disposition, urinary excretion and dosage regimen of subcutaneously administered levofloxacin in cross bred calves

The present study was conducted on five healthy male cross bred calves to study the kinetic dispositionand urinary excretion of levofloxacin following its single subcutaneous administration at the dose of4 mg.kg-1. The concentration of levofloxacin in plasma and urine samples was estimated by microbiologicalassay. Peak plasma level of levofloxacin (2.8 ± 0.3 μg.ml-1) was observed at 45 min and ...
